The aim of this study was to select an optimal method for the isolation of DNA from <em>Aspergillus flavus<\/em> suitable for PCR reaction. Four different methods were compared according to their efficiency in isolating pure DNA, their price and time consumption. DNA quantification and purity estimation were performed using the NanoDropTM 1000 UV\/VIS spectrophotometer and DNA integrity and PCR products were determined by gel-electrophoresis. DNA quantity ranged from 92.77 \u00b1 11.52 to 5477.4 \u00b1 22.03 ng\/\u00b5L, with A<sub>260<\/sub>\/<sub>280<\/sub> from 1.14 \u00b1 0.10 to 1.94 \u00b1 0.16, and A<sub>260<\/sub>\/<sub>230<\/sub> 0.37 \u00b1 0.05 to 1.91 \u00b1 0.17. There were also great differences in time consumption per sample, ranging from 1 hr 15 min to 7 hr 5 min. The determined costs per sample were ranging from 0.12 \u20ac to 2.29 \u20ac per sample.
